    Cytokine-mediated signal transduction in the JAK-STAT cascade requires the involvement of adaptor molecules. One such signal-transducing adaptor molecule contains an SH3 domain that is required for induction of MYC and cell growth. This protein binds to the SH3 domain of the signal-transducing adaptor molecule, and plays a critical role in cytokine-mediated signaling for MYC induction and cell cycle progression.     Encoded by human AMSH Gene, ubiquitously expressed 424-amino acid AMSH Protein contains two potential SH3-binding domains (PxxP motifs), a JAB1 subdomain homologous (JSH) region, and a putative nuclear localization signal. AMSH binds to the SH3 domain of STAM, which also contains an ITAM motif. The STAM/AMSH complex may signal MYC induction and cell cycle progression downstream of JAK3 and JAK2 after IL2 or GMCSF stimulation, respectively. (OMIM and NCI) (NCI Thesaurus)
